摘要:

视网膜新生血管的发生与发展涉及多种生长因子及其受体复杂的生物学过程,许多生长因子的受体均为蛋白酪氨酸激酶受体。蛋白酪氨酸激酶在新生血管的发生与发展中起非常重要的作用,生长因子生物学活性的表达需要与其靶细胞受体结合并激活受体酪氨酸激酶,引起其结构改变从而产生酶催化效应,引起生物学效应,促进新生血管形成。通过寻找选择性的酪氨酸酶抑制剂,来阻断或调控由于这些信号通路异常激活产生的疾病,是抗新生血管药物的研究策略。(国际眼科纵览, 2017,  41:   83-87)

Abstract:

The occurrence and development of angiogenesis involves the complex biological processes of multiple growth factors and their receptors. Many growth factor receptors are tyrosine kinase receptors. Protein tyrosine kinases play an important role in the occurrence and development of new blood vessels. The biological activity of growth factors needs to bind to its target cell receptors and activate receptor tyrosine kinase. The structure of  protein tyrosine kinase is changed to produce the enzyme catalytic effect, which can cause the biological effect, thus promoting the formation of new blood vessels. Searching for selective tyrosinase inhibitors to block or regulate the disease caused by abnormal activation of these pathways is a research strategy of anti angiogenesis drugs.
